目的探讨端粒酶hTERT和p27蛋白在子宫内膜癌发生、发展中的作用。方法应用链霉菌素蛋白-过氧化物酶免疫组织化学方法(S-P)对子宫内膜癌组织中端粒酶hTERT、p27蛋白表达进行检测,并与正常增生期子宫内膜组织中两者表达进行对比。结果端粒酶hTERT基因蛋白在子宫内膜癌组织的阳性表达率显著高于正常增生期子宫内膜组(P<0.05),其表达与年龄、组织学分级、临床分期、肌层浸润、及有无淋巴转移无关。正常增生期子宫内膜组p27蛋白的阳性表达率显著高于子宫内膜癌组(P<0.05)。p27蛋白在子宫内膜癌中的表达与组织学分级、临床分期、肌层浸润有关,而与年龄和有无淋巴转移无关。在子宫内膜癌中,端粒酶hTERT基因蛋白与p27蛋白的表达存在负相关关系。结论 hTERT高表达与p27蛋白的低表达协同作用促进内膜癌的发展,联合检测hTERT与p27蛋白有助于估计预后、指导治疗。
Objective To investigate the role of telomerase hTERT and p27 in the occurrence and development of endometrial carcinoma. Methods The expression of telomerase hTERT and p27 protein in endometrial carcinoma was detected by streptavidin-peroxidase immunohistochemistry (SP method), and the expression of telomerase hTERT and p27 in normal endometrium was detected Compared. Results The positive rate of telomerase hTERT protein expression in endometrial carcinoma was significantly higher than that in normal proliferative endometrium (P <0.05). The expression of telomerase hTERT protein was correlated with age, histological grade, clinical stage, myometrial invasion, No lymphatic metastasis has nothing to do. The positive expression rate of p27 protein in normal proliferative endometrium group was significantly higher than that in endometrial carcinoma group (P <0.05). The expression of p27 protein in endometrial carcinoma was related to histological grade, clinical stage and myometrial invasion, but not with age and lymphatic metastasis. In endometrial carcinoma, telomerase hTERT gene protein and p27 protein expression is negatively correlated. Conclusions The high expression of hTERT and the low expression of p27 protein promote the development of endometrial carcinoma. Combined detection of hTERT and p27 protein is helpful to predict the prognosis and guide the treatment.
