目的:检测核糖核酸酶T2(ribonuclease T2, RNaseT2)在雄性小鼠生殖系统和精子中的表达、定位及其对精子活动力及前向运动的影响。方法运用免疫荧光染色鉴定RNaseT2蛋白在小鼠生殖系统中的表达、定位及其与肌动蛋白(actin)在精子中的共定位关系,通过Real-time PCR检测RNaseT2 mRNA在小鼠生殖系统中的相对表达水平,借助计算机辅助精液分析(CASA)仪检测RNaseT2对小鼠精子的活动力、前向运动的影响。结果(1) RNaseT2蛋白及其mRNA在小鼠附睾头、体、尾组织以及输精管、前列腺及精囊腺分泌液中均有表达,而在睾丸组织中极低表达或未见表达。(2)在小鼠精子上,该蛋白定位于顶体、颈部、尾部鞭毛并与actin的表达定位具有一定的相似性。(3)体外纯化的RNaseT2蛋白可明显抑制小鼠精子的活动力及前向运动,且抑制具有时间及剂量依赖性。结论在雄性生殖系统中,RNaseT2蛋白作为精子运动的调节因子对精子的活动力及前向运动具有明显的抑制作用。“,”Objective To analyze the expression and distribution of RNase T2 in male mouse reproductive system and explore its effects on sperm motility. Methods The expression and localization of RNase T2 in the male mouse reproductive system and its co-localization with actin on mature spermatozoa were detected by indirect immunofluorescence analysis. The expression of RNase T2 in male mouse reproductive system was measured by real-time PCR. The effect of RNase T2 on mouse sperm motility and progressive motility were analyzed by a computer assisted semen analyzer (CASA). Results 1. RNase T2 was expressed in mouse epididymal caput, corpus, and cauda, and secretions of vas deferens, prostate and seminal vesicle, however no Rnase T2 was found in mouse testis. 2. RNase T2 was co-localized with actin on the sperm head, neck and the tail regions. 3. RNase T2 had an inhibitory effect on the sperm motility and progressive motility in a dose-dependent and time-dependent manners. Conclusion As a regulator for sperm motility, RNase T2 in male reproductive system has a definitely inhibitory effect on the motility and progressive motility of mouse spermatozoa.
