目的观察穿山龙总皂苷(Rhizoma Dioscoreae nipponicae,RDN)对体外白介素-1β(r IL-1β)诱导的成纤维样滑膜细胞(FLS)基质细胞衍生因子-1(SDF1)及IκB激酶(IKK)表达的影响。方法原代培养FLS,取第3代对数生长期的细胞将其分为正常组、模型组及给药组,模型组给予10μg/L r IL-1β诱导FLS增殖,给药组给予10μg/L r IL-1β及100μg/L穿山龙总皂苷共同孵育,正常组不予处理,孵育72 h后采用Western blot法测定各组大鼠FLS中SDF1及IKK(p-IKK)的表达水平。结果与正常组比较,模型组SDF1及p-IKK表达水平明显升高(P<0.01);与模型组比较,给药组FLS中SDF1及p-IKK表达水平明显降低(P<0.01)。结论穿山龙总皂苷可抑制SDF-1及IκB激酶的激活,可能通过对SDF-1的表达调控进而调控IKK的表达。
Objective To observe the effect of Rhizoma Dioscoreae nipponicae (RDN) on the expression of stromal cell derived factor-1 (SDF1) and IκB kinase (IKK) in fibroblast-like synoviocytes (FLS) induced by interleukin-1β Impact. Methods Primary cultured FLS cells were divided into normal group, model group and administration group at the third generation of logarithmic growth phase. The model group received 10 μg / L r IL-1β to induce FLS proliferation. The administration group received 10 μg / L r IL-1βand 100μg / L S. dauricii were incubated in normal group. The expression of SDF1 and IKK (p-IKK) in FLS of each group were detected by Western blot 72 hours after incubation. Results Compared with the normal group, the expression of SDF1 and p-IKK in the model group was significantly increased (P <0.01). Compared with the model group, the expressions of SDF1 and p-IKK in the FLS group were significantly decreased (P <0.01). Conclusion Scutellaria total saponin can inhibit the activation of SDF-1 and IκB kinase, which may regulate the expression of IKK by regulating the expression of SDF-1.
