目的炎症在慢性鼻-鼻窦炎伴鼻息肉(chronic rhinosinusitis with nasal polyps,CRSwNP)的发生发展过程中起着重要作用,并且炎症在CRSwNP的病理机制仍然不完全清楚。此研究主要观察清道夫受体CD36在慢性鼻-鼻窦炎伴鼻息肉的表达。方法使用逆转录酶聚合酶链反应(RT-PCR),Western blot免疫印迹方法检测来自合计43例慢性鼻-鼻窦炎(chronic rhinosinusitis,CRS)或伴息肉患者的手术标本CD36的mRNA和蛋白表达。进一步通过免疫组织化学和免疫荧光技术观察CD36表达与分布规律。结果我们的RT-PCR和Western blot免疫印迹分析的结果表明,与正常鼻黏膜组织相比,鼻息肉标本中CD36 mRNA和蛋白表达显著增强(P<0.01)。免疫组织化学结果显示,CD36弱表达于慢性鼻窦炎鼻黏膜组织,而鼻息肉组织中表达较强。CD36与血管内皮标记物CD31的双染荧光结果显示,CD36阳性的细胞与CD31的阳性共标。结论研究数据表明CD36在慢性鼻-鼻窦炎伴鼻息肉发生与发展过程可能起着重要作用。
The purpose of inflammation plays an important role in the development of chronic rhinosinusitis with nasal polyps (CRSwNP), and the pathological mechanism of inflammation in CRSwNP is still not fully understood. This study mainly observed the scavenger receptor CD36 expression in chronic rhinosinusitis with nasal polyps. Methods Reverse transcriptase polymerase chain reaction (RT-PCR) and Western blot were used to detect the mRNA and protein expression of CD36 from the total 43 cases of patients with chronic rhinosinusitis (CRS) or with polyps. Further observe the expression and distribution of CD36 by immunohistochemistry and immunofluorescence techniques. Results Our RT-PCR and Western blot results showed that the expression of CD36 mRNA and protein in nasal polyps was significantly increased (P <0.01) compared with normal nasal mucosa. Immunohistochemistry showed that CD36 was weakly expressed in nasal mucosa of chronic sinusitis and was strongly expressed in nasal polyps. Double-staining of CD36 with the vascular endothelial marker CD31 showed that CD36-positive cells were positive for CD31. Conclusion The data suggest that CD36 may play an important role in the pathogenesis and development of chronic rhinosinusitis with nasal polyps.
