染色体片段导入系是进行QTL精细定位、研究QTL互作和杂种优势机理的重要材料,分子标记辅助选择是染色体片段导入系创建中外源片段鉴定的重要手段。为了明确大白菜(Brassica campestris ssp.chinensis)—不结球白菜(Brassica campestris ssp.pekinensis)染色体导入系创建过程回交后代中外源染色体片段导入和丢失情况,以及外源片段对于大白菜表型性状的影响,本实验在对大白菜85-1和不结球白菜Jinglv No.2多态性In Del标记筛选的基础上,进一步对大白菜—不结球白菜BC_2F_1代进行了导入外源片段的In Del标记鉴定和表型分析。结果表明,供试的264个大白菜In Del标记,有186个在85-1和Jinglv No.2间表现出多态性,其中Jinglv No.2相对85-1特异的In Del标记有160个,分布于白菜(B.rapa)的10条染色体,每条染色体特异标记数为6~26个不等。从10条染色体不同位置挑选52个In Del标记用以鉴定大白菜—不结球白菜BC_2F_1代群体,其中50个In Del标记在196个BC_2F_1植株中以不同频率鉴定出来,含有8个外源片段的植株数最多。通过GGT(Graphical Geno Typing)软件分析,筛选出导入外源片段较少且携带有50个In Del标记的外源片段的BC_2F_1植株40株,可进一步用于单片段导入系的建立。进一步表型观测表明,BC_2F_1代单株在叶片形状、大小、颜色及抽薹性、花色等性状上表现了分离。本研究结果为整套大白菜—不结球白菜染色体片段导入系的创建提供了技术支持,并为研究不同外源片段的导入对植株表型变异影响提供了材料。
Chromosome fragment introduction is an important material for QTL fine mapping, QTL interaction and heterosis mechanism. Molecular marker-assisted selection is an important method for identification of foreign fragments in chromosome segment introduction. In order to clarify the introduction and loss of exogenous chromosome segments in backcross progenies of Brassica campestris ssp.chinensis-Brassica campestris ssp.pekinensis chromosome inbred lines and the effects of exogenous fragments on the phenotypic traits of Chinese cabbage In this study, based on the screening of In Del markers of Chinese cabbage 85-1 and Jinglv No.2 of Chinese cabbage, the BC 2 F 1 generation of Chinese cabbage-non-heading Chinese cabbage was further introduced with foreign DNA In Del marker identification and phenotypic analysis. The results showed that there were 186 In Del markers in the tested Chinese cabbage, 186 showed polymorphism between 85-1 and Jinglv No.2, of which 160 were in Jinglv No.2 relative to 85-1 specific In Del , Distributed in cabbage (B.rapa) 10 chromosomes, each chromosome-specific markers ranging from 6 to 26. Fifty-two In Del markers were selected from 10 chromosomes to identify the BC_2F_1 population of Chinese cabbage-non-heading Chinese cabbage. Fifty In Del markers were identified in 196 BC_2F_1 plants at different frequencies and contained 8 exogenous fragments The largest number of plants. The results of GGT (Graphical Genotyping) software showed that 40 BC_2F_1 plants with fewer foreign DNA fragments and 50 In Del-labeled exogenous DNA fragments were obtained. Further phenotypic observations showed that BC2F1 plants showed segregation in leaf shape, size, color and bolting, flower color and other traits. The results of this study provided technical support for the establishment of the whole genome-introduced chromosome fragment in Chinese cabbage and provided the material for studying the effects of different foreign fragments on plant phenotypic variation.
