本文用免疫组化技术研究32例乳腺癌中增殖细胞核抗原(PCNA)的表达情况,并联系其它预后因素及随访结果,探讨其预后意义。 1 材料与方法 对病理档案中资料比较完整的女性乳腺癌病例进行回顾性复习,并按美国最近颁布的乳腺标本检查记录的实用方案重新进行分类和分级,同时根据病历提供的线索用电话、通讯等方式进行随访,共获得完整资料32例。所有病理均用10%福尔马林固定,石蜡包埋,苏木素伊红染色,光镜观察。然后选择癌组织作免疫化标记(ABC法),所用PCNA抗体为鼠抗人单克隆抗体(PCIO),工作浓度1:20,DAB显色,苏木素复染,按规定程序操作。用人肝组织作阴性对照。 标记结果,以细胞核内出现棕黄色颗粒为阳性,不见或偶见阳性颗粒者为阴性。PCNA在癌细胞中的阳性率占25%计为1分,26%~50%计为2分,51%~75%为3分,超过75%者计为4分,视为高度表达。
In this paper, the expression of proliferating cell nuclear antigen (PCNA) in 32 cases of breast cancer was studied by immunohistochemical technique, and other prognostic factors and follow-up results were also discussed to explore its prognostic significance. 1 Materials and Methods Retrospective review of female breast cancer cases with relatively complete data in the pathological archives, and reclassification and classification according to the practical program for the examination of breast specimens recently enacted in the United States. At the same time, according to the clues provided by medical records, telephone and communication are used. Follow-up visits were conducted in a total of 32 cases. All pathologies were fixed with 10% formalin, embedded in paraffin, stained with hematoxylin and eosin, and observed under a light microscope. Then, the cancerous tissue was selected for immunolabeling (ABC method). The PCNA antibody used was mouse anti-human monoclonal antibody (PCIO), working concentration 1:20, DAB coloration, hematoxylin counterstain, and procedures were followed. Human liver tissue was used as a negative control. As a result of the labeling, brownish yellow particles were found to be positive in the nucleus, and negative or negative particles were found to be negative. The positive rate of PCNA in cancer cells is 2%, 1 point, 26% to 50%, 2 points, 51% to 75%, 3 points, and 75% or more, which is considered highly expressed.
