目的 :探寻一种快速简便的肝癌细胞体外药敏试验方法。方法 :选用白血病 K562 细胞株和鼠肝癌 RH— 35细胞株对 MTT快速比色法试验条件进行初步探讨 ,并对 2 0例原发性肝癌细胞进行药敏试验。结果 :MTT甲簪产物最大吸收峰波长为 51 5nm,在细胞数为 0 .5~ 4× 1 0 5/ ml范围内 ,细胞密度与甲簪产量成直线关系 ,γ=0 .9998;2 0例肝癌细胞药敏试验成功率为 80 % ,敏感率为 2 2 .2 %~ 75.0 %。结论 :MTT法能反应出癌细胞的存活率 ,且时间短 ,操作简便 ,重复性好 ,成功率高
Objective: To explore a rapid and simple method for the in vitro drug susceptibility testing of hepatoma cells. METHODS: A preliminary study was conducted on the MTT rapid colorimetric assay conditions in leukemia K562 cell line and rat liver cancer RH-35 cell line, and drug susceptibility testing was performed on 20 primary hepatoma cells. RESULTS: The maximum absorption peak wavelength of MTT formazan product was 51 5 nm. In the cell number range of 0.5 to 4×10 5 / ml, the cell density was linear to the production of formazan, γ=0.9998; 2 0 The success rate of the drug susceptibility test of hepatoma cells was 80%, and the sensitivity rate was 22.2% to 75.0 %. Conclusion: The MTT method can reflect the survival rate of cancer cells with short time, easy operation, good repeatability, and high success rate.
