本研究利用SSR标记技术筛选合适的引物,对辽宁省近年备案的34个大白菜品种进行DNA指纹分析,构建其SSR指纹图谱数据库。首先利用8份来自国内外不同生态条件、植物学性状差异较大的大白菜品种,通过对1 200余对SSR引物的两次筛选,得到40对扩增条带清晰、多态性好、稳定性高且近似均匀分布在白菜10条染色体上的引物。通过选取的40对SSR引物对34份通过备案的大白菜品种进行扩增分析,最终利用Cnu-m132a等7对引物构建了上述34份大白菜品种的指纹图谱。本研究构建的SSR指纹图谱数据库,为建立辽宁大白菜品种鉴定技术体系提供参考依据。
In this study, SSR markers were used to screen the suitable primers, DNA fingerprinting was performed on 34 Chinese cabbage cultivated in Liaoning Province in recent years, and its SSR fingerprinting database was constructed. Firstly, using eight Chinese cabbage cultivars with different ecological conditions and different botanical traits from home and abroad, 40 pairs of SSR primers were screened by double screening of more than 1 200 pairs of SSR primers, showing good polymorphism and stability Highly uniform and approximately uniformly distributed in cabbage 10 chromosome primers. A total of 34 SSR primers were used to amplify the reported Chinese cabbage varieties. The fingerprints of the above 34 Chinese cabbage varieties were constructed using 7 pairs of primers, such as Cnu-m132a. The SSR fingerprinting database constructed in this study provides a reference for the establishment of Liaoning cabbage variety identification technology system.
