目的:探讨赤芍药提取物对高脂喂养兔颈动脉球囊损伤后血管内膜增殖与还原型烟酰胺腺嘌呤二核苷酸磷酸(NADPH)氧化酶亚单位p22phox及单核细胞趋化蛋白-1(MCP-1)的影响。方法:采用Clowes方法建立兔颈总动脉球囊损伤模型,并给予赤芍药提取物。免疫组化染色测定兔颈动脉新生内膜成分、增殖细胞核抗原(proliferat ingcellnuclearantigen,PCNA)及巨噬细胞表达,血管特殊染色观察1型胶原增生,图像分析系统测定新生内膜面积;原位杂交染色及逆转录聚合酶链反应检测增殖血管内膜的NADPH Oxidase p22phox及MCP-1 mRNA水平。结果:赤芍药提取物可减少PCNA阳性细胞数,抑制型胶原增生,减轻新生内膜形成;显著降低NADPH Oxidase p22phox及MCP-1mRNA表达。结论:赤芍药抑制血管内膜增殖程度,其机制可能与抗氧化应激及抗血管内皮炎症反应有关。
OBJECTIVE: To investigate the effect of Paeoniae Radix extract on the intimal hyperplasia and the reduced nicotinamide adenine dinucleotide phosphate (NADPH) oxidase subunit p22phox and monocyte chemoattractant protein in carotid artery after balloon injury in rabbits fed with high fat diet. Effect of 1 (MCP-1). METHODS: The Clowes method was used to establish the balloon injury model of common carotid artery in rabbits, and the extract of Radix Paeoniae Rubra was extracted. Immunohistochemical staining was used to determine the neointimal components of carotid artery in rabbits, proliferat ing cell nuclear antigen (PCNA) and macrophage expression. Type 1 collagen hyperplasia was observed by special staining of blood vessels. Image analysis system was used to measure neointima area; in situ hybridization staining. The mRNA levels of NADPH Oxidase p22phox and MCP-1 in proliferative intima were detected by reverse transcription polymerase chain reaction. RESULTS: Radix Paeoniae Rubra could reduce the number of PCNA-positive cells, inhibit collagen hyperplasia, and reduce the formation of neointima; it significantly reduced the expression of NADPH Oxidase p22phox and MCP-1 mRNA. CONCLUSION: The inhibitory effect of Radix Paeoniae Rubra on intimal hyperplasia may be related to antioxidative stress and anti-vascular endothelial inflammation.
