采用无菌培养方法在琼脂固体培养基上对龙胆种子,离体器官茎尖及茎段进行培养,成功地培养出了龙胆试管苗。培养四个月的龙胆种子可发育成株高3~4cm,具4~6条或更多不定根及3~4个节的植株。培养五个月的试管实生苗其单株根鲜重为栽培条件下一年生实生苗根重(0.05克)的5.6倍。茎段。茎尖可在离体培养后30及49天生根发育成完整植株。不同种龙胆对pH值适应范围有差别,培养基pH值以及生长激素对试管苗生长的影响明显。应用试管苗加快了龙胆育苗速度在生产上有实用价值,并为龙胆育苗工厂化提供依据。
Aseptic culture methods were used to culture gentian seeds, shoot tips and stems of in vitro organs on agar solid medium and successfully cultured gentian test tube seedlings. The gentian seeds cultivated for four months can grow into plants with 3 ~ 4 cm in height with 4 ~ 6 or more adventitious roots and 3 ~ 4 nodes. The five-month-old test-tube seedlings had 5.6-fold root fresh weight per plant under the cultivation conditions. Stems. The shoot tips developed into complete plants 30 and 49 days after in vitro culture. Different genus gentian to adapt to the pH range, media pH value and growth hormone on the growth of test tube significantly. Application of tube seedlings to speed up the gentian breeding speed in the production of practical value, and provide the basis for gentian nursery factory.
