目的分析人肠道病毒71型(EV-A71)感染神经母细胞瘤来源的SK-N-SH细胞的microRNA(miRNA)表达谱差异。方法用EV-A71病毒感染人SK-N-SH细胞,6h后收获总RNA,基因芯片检测感染和未感染EV-A71的神经细胞miRNA表达谱,并用实时荧光定量PCR方法 (qRT-PCR)进行验证。结果比较EV-A71感染与未感染的SK-N-SH细胞miRNA的表达谱,SK-N-SH细胞被EV-A71感染后有26种miRNA表达显著上调,40种miRNA表达显著下调。结论 EV-A71感染显著改变了SK-N-SH神经细胞的miRNA表达谱,可作为EV-A71感染机制深入研究的实验基础。
Objective To analyze the difference of microRNA (miRNA) expression profiles of SK-N-SH cells derived from human enterovirus 71 (EV-A71) infected neuroblastoma. Methods Human SK-N-SH cells were infected with EV-A71 virus and total RNA was harvested 6 hours later. The miRNA expression profiles of infected and non-infected EV-A71 neurons were detected by real-time PCR and qRT-PCR verification. Results The expression profiles of miRNAs in EV-A71 infected and uninfected SK-N-SH cells were compared. 26 miRNAs were significantly up-regulated and 40 miRNAs were significantly down-regulated in SK-N-SH cells infected with EV-A71. Conclusion EV-A71 infection significantly altered the miRNA expression profile of SK-N-SH neurons and could serve as an experimental basis for in-depth studies on the mechanism of EV-A71 infection.
