本试验从70条ISSR引物中筛选出15条多态性好的ISSR引物,对10份原产自云南的彩色马铃薯材料进行遗传多样性分析和ISSR指纹图谱构建。结果表明,15条引物共扩增到101条清晰条带,89个多态性条带,多态性条带百分率为89.24%,引物平均多态性位点5.93,平均多态性信息量0.823。聚类分析可将10份彩色马铃薯划分为4类,其中S03-2677和S03-2685的遗传相似系数最大,而紫云1号和S06-277的遗传相似系数最小。同时筛选到1条核心引物AW67776,绘制了10份彩色马铃薯品种的指纹图谱。结果表明ISSR标记能够较准确地按亲缘关系划分彩色马铃薯品种,可为我国彩色马铃薯的品种选育提供参考。
Fifteen ISSR primers with good polymorphism were screened from 70 ISSR primers. Genetic diversity and ISSR fingerprinting of 10 potato cultivars originated from Yunnan were studied. The results showed that a total of 101 clear bands and 89 polymorphic bands were amplified by 15 primers, the percentage of polymorphic bands was 89.24%, the average polymorphic locus of primers was 5.93, the average amount of polymorphic information was 0.823 . Cluster analysis could divide 10 color potatoes into 4 categories, of which the genetic similarity coefficients of S03-2677 and S03-2685 were the largest, while the genetic similarities of Ziyun 1 and S06-277 were the smallest. At the same time, a core primer AW67776 was screened and the fingerprints of 10 color potato cultivars were drawn. The results showed that the ISSR markers could be used to classify color potato varieties more accurately and provide reference for the breeding of color potato in China.
