目的观察抗疏健骨颗粒对去卵巢大鼠骨组织形态学的影响,探讨抗疏健骨颗粒防治骨质疏松症的作用机理。方法切除大鼠双侧卵巢诱发大鼠骨质疏松症模型,连续灌胃给药12 W,各组动物于处死前15 d和前5 d分别进行四环素荧光标记,处死动物后取右侧胫骨,制作切片,在荧光显微镜下观察骨小梁形态;采用图象分析系统进行骨组织形态参数计量,并与手术组、模型组和尼尔雌醇组进行比较。结果与模型组比较,抗疏健骨颗粒大、中剂量组骨小梁显著增加,骨小梁排列较紧密,骨小粱间可见网状连接,骨小梁厚度增加;与假手术组比,模型组较骨小梁体积百分比(TBV%)明显下降(P<0.01);尼尔雌醇组、抗疏健骨颗粒大、中与模型组比较,TBV明显增加(P<0.01),并呈一定的剂量依赖性。模型组较假手术组骨吸收参数(TRS)、骨形成参数(TFS)明显增加(P<0.01);与模型组比较抗疏健骨颗粒大、中均能降低骨吸收参数TRS、骨形成参数(TFS)、MAR、mAR和osw(P<0.01或P<0.05)。结论抗疏健骨颗粒能改善骨形态计量学参数,对骨质疏松症具有一定的治疗作用。
Objective To observe the effect of Shuzheng Jiangu Granule on bone histomorphology in ovariectomized rats and to explore the mechanism of Shuzheng Jiangu Granule in preventing and treating osteoporosis. Methods The rat model of osteoporosis induced by bilateral ovary was excised and the rats were intragastrically administrated for 12 weeks. The animals in each group were labeled with tetracycline 15 days before and 5 days before sacrifice. After the animals were sacrificed, the right tibia, The sections of the trabecular bone were observed under a fluorescence microscope. The morphological parameters of the bone tissue were measured by image analysis system and compared with the surgery group, the model group and the nilestriol group. Results Compared with the model group, trabecular bone was significantly increased in the groups of large dose and middle dose of anti-bone-sparing particles, trabecular arranged in close proximity, and the trabecular meshwork connections were found between the trabecular bone and the trabecular bone. Compared with the model group, TBV significantly increased (P <0.01) in the model group and TBV% (P <0.01) Certain dose-dependent. The bone resorption parameter (TRS) and bone formation parameter (TFS) in the model group were significantly higher than those in the sham operation group (P <0.01). Compared with the model group, (TFS), MAR, mAR and osw (P <0.01 or P <0.05). Conclusion Anti-Shuangjiangu granules can improve bone morphometry parameters, and have a certain therapeutic effect on osteoporosis.
