目的:探讨坐骨神经损伤后脊髓前角细胞BDNF表达变化及外源性NGF与BDNF表达的关系。方法:采用大鼠坐骨神经损伤动物模型,分为药物组、对照组,每只大鼠又分为损伤例组(L组)和健侧组(R组)两个亚组。用原位杂交、免疫级化技术检测BDNF的表达水平,现察各组在1d、7d、14d、21d及28d时的变化。用计算机图像自动处理系统进行定量分析研究。结果:药物组损伤侧脊髓前角细胞中BDNF表达水平除1d,7d组外其余均多于对照组(P<0.05);而两组健侧脊髓前角细胞中BDNF表达水平无统计学意义(P>0.05),坐骨神经损伤后损伤侧脊髓前角细胞中BDNF表达水平在最初的3周内逐渐下降,然后开始恢复,但至第4周尚未恢复到开始时的水平;而两组健侧BDNF表达水平则无明显差异(P>0.05)。结论:外源性NGF能促进坐骨神经损伤后损伤侧脊髓前角细胞中BDNF表达水平的增加;而对健侧BDNF表达水平无明显作用。
Objective: To investigate the changes of BDNF expression in spinal cord anterior horn cells and the relationship between exogenous NGF and BDNF expression after sciatic nerve injury. Methods: The animal models of sciatic nerve injury in rats were divided into drug group and control group. Each rat was divided into two subgroups: injury group (L group) and contralateral group (R group). The expression of BDNF was detected by in situ hybridization and immunohistochemistry. The changes of BDNF in each group were observed at 1d, 7d, 14d, 21d and 28d. Computer image automatic processing system for quantitative analysis. Results: Compared with the control group, BDNF expression in the anterior horn of spinal cord in the injury group was higher than that in the control group (P <0.05), but there was no significant difference in BDNF expression in the anterior horn cells P> 0.05). After sciatic nerve injury, the expression of BDNF in the injured spinal cord anterior horn cells gradually decreased in the first 3 weeks and then began to recover, but it did not recover to the initial level in the 4th week. However, BDNF There was no significant difference in the expression level (P> 0.05). CONCLUSION: Exogenous NGF can promote the increase of BDNF expression in injured spinal cord anterior horn cells after sciatic nerve injury, but has no effect on the expression of BDNF.
