目的探讨重组人肝再生增强因子(rocombinant human augmenter of liver regeneration,rhALR)对5/6肾切除所致慢性肾衰竭大鼠肾功能的保护作用。方法将雄性SD大鼠分为假手术组、对照组及rhALR组,以rhALR对5/6肾切除所致慢性肾衰竭大鼠进行治疗,比较各组大鼠血清尿素氮(BUN)、肌酐(Scr)及肾脏病理改变各项指标。结果5/6肾切除后,大鼠血中Scr及BUN升高,病理学检查见肾间质纤维化,慢性肾衰竭大鼠模型制备成功。给予rhALR能降低慢性肾衰竭大鼠血中Scr及BUN水平,减少肾间质纤维化面积。结论rhALR可有效降低5/6肾切除所致慢性肾衰竭大鼠的Scr及BUN水平,改善肾脏病理改变,延缓慢性肾衰竭进展,保护残肾功能。
Objective To investigate the protective effect of recombinant human augmenter of liver regeneration (rhALR) on renal function in rats with chronic renal failure induced by 5/6 nephrectomy. Methods Male Sprague-Dawley rats were randomly divided into sham operation group, control group and rhALR group. Rats in chronic renal failure caused by 5/6 nephrectomy were treated with rhALR. The levels of serum urea nitrogen (BUN) and creatinine Scr) and renal pathological changes of the indicators. Results After 5/6 nephrectomy, the levels of Scr and BUN in rat blood increased. Renal interstitial fibrosis was found in pathological examination. The rat model of chronic renal failure was successfully established. Administration of rhALR decreased the levels of Scr and BUN in the blood and decreased the area of renal interstitial fibrosis in rats with chronic renal failure. Conclusion rhALR can effectively reduce Scr and BUN levels in 5/6 nephrectomized rats with chronic renal failure, improve renal pathological changes, delay the progression of chronic renal failure, and protect residual renal function.
