目的:研究干扰素-α(IFN-α)对肝纤维化大鼠肝星状细胞凋亡的影响,观察IFN-α治疗后肝纤维化程度的变化.方法:用四氯化碳(CCl4)复制肝纤维化模型大鼠32只.造模8wk后处死6只作为模型对照组(E组),另外26只分为B、C两组,B组14只,给予IFN-α0.1MU/100g皮下注射8wk,C组12只,用等量的生理盐水注射8wk.治疗结束时用TUNEL法检测各组肝星状细胞凋亡情况,测定血清Ⅲ型前胶原水平、免疫组化检测Desmin、α-SMA、I型胶原、TGF-β1等指标.结果:B组较其他组肝组织学改善明显,肝纤维化程度减轻,I型胶原、TGF-β1的显色指数降低,肝星状细胞明显减少,Desmin与TUNEL双染色均阳性的细胞增多,凋亡指数增加,与C组相比,有显著差异.结论:IFN-α诱导HSC发生凋亡可能是其抗肝纤维化的机制之一.
Objective: To investigate the effect of interferon-α (IFN-α) on hepatic stellate cell apoptosis in rats with hepatic fibrosis and to observe the changes of liver fibrosis after IFN-α treatment.Methods: CCl4 Sixty-two rats were randomly divided into B group and C group, and 14 rats in B group were given IFN-α0.1MU / 100g Subcutaneous injection of 8wk, C group 12, with the same amount of saline injection 8wk.At the end of treatment by TUNEL assay of each group of hepatic stellate cell apoptosis, determination of serum type Ⅲ procollagen levels, immunohistochemical detection of Desmin, α SMA, type I collagen, TGF-β1 and other indicators.Results: Compared with the other groups, B group improved significantly liver histology, reduced the degree of liver fibrosis, type I collagen, TGF-β1 decreased the color index, hepatic stellate cells were significantly Decreased, Desmin and TUNEL double staining positive cells increased, apoptosis index increased, compared with the C group, there are significant differences.Conclusion: IFN-α-induced HSC apoptosis may be one of the mechanisms of its anti-liver fibrosis.
