目的:探讨茵芍散含药血清对大鼠肝星状细胞(HSC)增殖作用的影响及其机制。方法:40只Wistar大鼠分别给予中药高、中、低剂量、0.9%氯化钠溶液、己酮可可碱灌胃,喂养1周后,腹主动脉采血,制备药物血清。设中药高、中、低剂量组,阴性对照组,阳性对照组及空白组,药物血清干预HSC-T6后,运用流式细胞术、吖啶橙/溴化乙锭(AO-EB)染色观察细胞增殖、分析周期情况,并分别通过实时荧光定量PCR、Western blot检测β-catenin m RNA及蛋白表达情况。结果:与对照组比较,茵芍散能有效抑制HSC-T6增殖,下调β-catenin m RNA及蛋白表达(P<0.05),且具有量-效关系。结论:茵芍散可抑制HSC增殖,阻断Wnt/β-catenin信号通路对HSC的激活可能是其治疗肝纤维化的作用机制。
Objective: To investigate the effect and mechanism of Yinshao San containing serum on the proliferation of rat hepatic stellate cells (HSC). Methods: Forty Wistar rats were given high, medium and low doses of Chinese medicine, 0.9% sodium chloride solution and pentoxifylline respectively. After feeding for one week, abdominal aorta blood was collected to prepare drug serum. HSC-T6 was treated with high, medium and low dose of Chinese herbal medicine, negative control group, positive control group, blank control group and serum of drug-treated HSC-T6 cells by flow cytometry and AO-EB staining Cell proliferation and analysis of cycle conditions, and β-catenin m RNA and protein expression were detected by real-time fluorescence quantitative PCR and Western blot respectively. Results: Compared with the control group, YinShao Powder can effectively inhibit the proliferation of HSC-T6 and down-regulate the expression of β-catenin mRNA and protein (P <0.05), and has a dose-response relationship. CONCLUSION: YinShao powder can inhibit the proliferation of HSC. Blocking the activation of HSC by Wnt / β-catenin signaling pathway may be the mechanism of its action in treating hepatic fibrosis.
