目的:建立HPLC法同时测定咳喘平散中芥子碱硫氰酸盐、细辛脂素、五味子甲素、五味子乙素含量。方法:采用C_(18)(4.6 mm×250 mm,5μm)色谱柱,流动相为乙腈-0.08 mol·L~(-1)磷酸二氢钠水溶液,梯度洗脱,流速1.0 m L·min~(-1),柱温30℃,检测波长为286 nm(芥子碱硫氰酸盐、细辛脂素)、220 nm(五味子甲素、五味子乙素)。结果:芥子碱硫氰酸盐、细辛脂素、五味子甲素、五味子乙素线性范围为0.238~1.789、0.061~0.457、0.071~0.533和0.069~0.521μg;平均回收率为98.7%~101.8%,RSD为1.6%~1.8%。3批样品中芥子碱硫氰酸盐、细辛脂素、五味子甲素和五味子乙素4个成分的含量分别为1.508~1.566、0.470~0.497、0.398~0.420、0.470~0.553 mg·g~(-1)。结论:本法可为咳喘平散及其类似组方制剂的质量评价提供研究基础。
OBJECTIVE: To establish a HPLC method for simultaneous determination of sinapine thiocyanate, sino-octylin, schisandrin and schisandrin B in Kechuanping Powder. Methods: The mobile phase consisted of C18 (4.6 mm × 250 mm, 5 μm) column with a mobile phase of acetonitrile - 0.08 mol·L -1 sodium dihydrogenphosphate aqueous solution with gradient elution at a flow rate of 1.0 m L · min ~ (-1), the column temperature was 30 ℃, the detection wavelength was 286 nm (sinapine thiocyanate, zintolipin), 220 nm (schisandrin, Schisandrin B). Results: The linear ranges of sinapine thiocyanate, sinoaicin, schisandrin and schisandrin were 0.238 ~ 1.789, 0.061 ~ 0.457, 0.071 ~ 0.533 and 0.069 ~ 0.521μg respectively.The average recoveries were 98.7% ~ 101.8% , RSD was 1.6% ~ 1.8%. The contents of 4 components of sinapine thiocyanide, sino-azoin, schisandrin and schisandrin B in three batches of samples were 1.508-1.566, 0.470-0.497, 0.398-0.420, 0.470-0.553 mg · g ~ (-1) -1). Conclusion: This method can provide the basis for the quality evaluation of Kechuan Ping San and its similar prescriptions.
