目的 探讨Notch-2蛋白和Numb蛋白在甲状腺乳头状癌(papillary thyroid carcinoma,PTC)组织中的表达及其临床意义.方法 回顾性收集2014年3月至2017年3月期间于内蒙古民族大学附属医院行手术切除的50例PTC患者的癌组织及其癌旁组织标本,采用免疫组织化学染色方法检测癌组织及其癌旁组织中Notch-2蛋白和Numb蛋白的表达.结果 ①癌组织和癌旁组织中Notch-2蛋白和Numb蛋白的表达:癌组织中Notch-2蛋白的表达阳性率为82.00% (41/50),高于癌旁组织的18.00% (9/50),差异有统计学意义(x2=40.960,P<0.001);癌组织中Numb蛋白的表达阳性率为66.00% (33/50),高于癌旁组织的0(0/50),差异有统计学意义(x2=49.254,P<0.001).②癌组织中Notch-2蛋白和Numb蛋白表达的相关性:癌组织中Notch-2蛋白与Numb蛋白的表达呈正相关关系(rs=0.323,P=0.022).③癌组织中Notch-2蛋白和Numb蛋白的表达与PTC患者临床病理学特征的关系:癌组织中Notch-2蛋白的表达与患者的性别、年龄、肿瘤直径、包膜浸润、颈部淋巴结转移及TNM分期均无关(P>0.05);癌组织中Numb蛋白的表达与患者的性别、年龄、肿瘤直径和包膜浸润均无关(P>0.05),而与颈部淋巴结转移和TNM分期均有关(P<0.05),无颈部淋巴结转移组患者的表达阳性率高于发生颈部淋巴结转移组,Ⅰ期+Ⅱ期组患者的表达阳性率高于Ⅲ期+Ⅳ期组.结论 PTC组织中Notch-2蛋白和Numb蛋白的表达阳性率均较癌旁组织高,且两者在癌组织中的表达存在正相关关系,提示两者可能在PTC的病程进展过程中存在协同作用.“,”Objective To investigate the expressions and clinical significance of Notch-2 protein and Numb protein in papillary thyroid carcinoma (PTC).Methods PTC tissues and its para-cancerous tissues of 50 patients with PTC who treated in The Affiliated Hospital of Inner Mongolia University for Nationalities from Mar.2014 to Mar.2017 were retrospectively collectied,to detect the expressions of Notch-2 protein and Numb protein by immunohistocheumical method.Results ① Expressions of Notch-2 protein and Numb protein in PTC tissues and para-cancerous tissues:the positive-expression rate of Notch-2 protein in PTC tissues was 82.00% (41/50),which was higher than that of paracancerous tissues [18.00% (9/50)],the difference was statistically significant (x2=40.960,P<0.001).The positiveexpression rate of Numb protein in PTC tissues was 66.00% (33/50),which was higher than that of para-cancerous tissues [0 (0/50)],the difference was statistically significant (x2=49.254,P<0.001).② The relationship between expression of Notch-2 protein and expression of Numb protein in PTC tissues:there was a positive correlation between expressions of Notch-2 protein and Numb protein in PTC tissues (r,=0.323,P=0.022).③ The relationship between expressions of Notch-2 protein and Numb protein in PTC tissues and clinicopathological features of the PTC patients:the expression of Notch-2 protein in PTC tissues was not significantly correlated with gender,age,tumor diameter,capsule infiltration,cervical lymph node metastasis,and TNM staging (P>0.05).The expression of Numb protein in PTC tissues was not significantly correlated with gender,age,tumor diameter,and capsule infiltration (P>0.05),but was significantly correlated with cervical lymph node metastasis and TNM staging (P<0.05),the positive rates of Numb protein in patients of staging Ⅲ + Ⅳ group and cervical lymph node metastasis group were lower than those of patients in staging Ⅰ + Ⅱ group and non-cervical lymph node metastasis group.Conclusion The positive-expression rate of Notch-2 protein and Numb protein in PTC tissues are higher than those of para-cancerous tissues,and there is a positive correlation between them in PTC tissues,suggesting that there may be a synergistic effect in the course of PTC progression.
