[目的]探讨氢醌(hydroquinone,HQ)对细胞周期的影响及其相关的调控机制。[方法]磷酸盐缓冲液(PBS)处理人类淋巴母细胞(TK6细胞)为对照组,20.0μmol/L HQ为处理组,48 h后通过碘化丙啶(PI)标记的流式细胞术检测细胞周期分布情况,用实时荧光定量聚合酶链反应(qRT-PCR)检测PTEN(抑癌基因)、p53基因mRNA表达的改变,流式细胞荧光标记法检测Rb蛋白的表达。[结果]与对照组比较,处理组G1期细胞比例明显增加,S期细胞比例明显减少,差异均有统计学意义(P<0.05);HQ作用于TK6细胞后上调了Rb的表达,下调了p53的表达,其差异均有统计学意义(P<0.05),而对PTEN的表达没有明显影响。[结论]HQ可能通过上调Rb的表达使细胞阻滞于G1期。
[Objective] To investigate the effect of hydroquinone (HQ) on cell cycle and its related regulatory mechanism. [Method] Human lymphoblasts (TK6 cells) were treated with phosphate buffered saline (PBS) in the control group and treated with 20.0 μmol / L HQ for 48 h and then detected by flow cytometry with propidium iodide (PI) The expression of PTEN (tumor suppressor gene) and p53 gene mRNA was detected by qRT-PCR, and the expression of Rb protein was detected by flow cytometry. [Results] Compared with the control group, the proportion of cells in G1 phase increased significantly and the proportion of cells in S phase decreased significantly (P <0.05). After treated with HQ, the expression of Rb was up-regulated in TK6 cells and down-regulated p53, the difference was statistically significant (P <0.05), but no significant effect on the expression of PTEN. [Conclusion] HQ may arrest cells in G1 phase by up-regulating Rb expression.
