本实验采用SDS-EDTA精子核解聚法,显示赖氨酸组蛋白的苯胺兰染色法以及磷钨酸(PTA)电镜细胞化学法对习惯性流产患者配偶射出精子的核成熟状况进行了研究。实验结果表明:与正常生育力男性的精子相比,习惯性流产患者配偶的精子核,抗SDS-EDTA解聚能力较低,苯胺兰染色阳性率和PTA染色阳性率较高。本实验结果提示某些习惯性流产患者配偶的精子核的稳定性较低,这可能与核蛋白组型转换异常有关,而核未完全成熟的精子可能是造成某些习惯性流产的病因之一。
In this study, SDS-EDTA sperm nuclear disaggregation method was used to study the nuclear maturation status of the spouses who had the habitual abortion due to the aniline staining of lysine histone and the electron microscopy of phosphotungstic acid (PTA). The experimental results show that compared with sperms of normal fertility males, sperm nucleus and anti-SDS-EDTA disaggregation ability of spouses of habitual abortion patients are lower than those of normal fertility males, and the positive rates of aniline blue staining and PTA staining are higher. Our results suggest that spouses in some habitual abortion patients with lower stability of the sperm nucleus, which may be abnormal nuclear proteomics conversion, and nuclear incomplete mature sperm may be caused by some of the causes of habitual abortion .
