目的:观察天升元口服液对免疫介导的再生障碍性贫血(immune-mediated aplastic anemia,IAA)模型小鼠外周血T淋巴细胞异常和正负造血调控因子水平的影响,以探讨其作用机制。方法:给予IAA模型小鼠天升元口服液(含生药6.2,12.4 g.kg-1.d-1)ig,14 d后,通过流式细胞术检测外周血CD4+,CD8+及其比值,并结合荧光免疫分析法检测外周血中造血正负调控因子(GM-CSF,TNF-α)的含量。结果:天升元口服液高剂量组CD8+及CD4+/CD8+分别为(15.89±8.32)%和(2.34±1.10),(n=7),模型对照组CD8+及CD4+/CD8+分别为(35.13±13.13)%和(0.92±0.33)(n=12),组间有非常显著差异(P<0.01~0.001);同时,天生元口服液高剂量组的GM-CSF含量(1 400±670)ng.L-1,(n=10),也显著高于模型对照组(665±276)ng.L-1,(n=12,P<0.01)。此外,TNF-α有一定程度的降低。结论:天升元口服液通过降低CD8+T淋巴细胞的产生,提高CD4+/CD8+比值,促进T淋巴细胞亚群的平衡,同时促进造血正调控因子(GM-CSF)的产生,从而呈现对IAA模型小鼠的治疗作用。
OBJECTIVE: To observe the effect of Tianshengyuan oral liquid on T-lymphocyte abnormality and the positive and negative hematopoietic regulatory factors in peripheral blood of immune-mediated aplastic anemia (IAA) model mice to explore its mechanism of action . Results: The levels of CD8 + and CD4 + / CD8 + of TSP group were (15.89 ± 8.32)% and (2.34 ± 1.10), respectively, and the CD8 + and CD4 + / CD8 + of the model control group were (35.13 ± 13.13 ) And (0.92 ± 0.33) (n = 12), respectively. There was a significant difference between the two groups (P <0.01-0.001). Meanwhile, the GM-CSF content of the high dose of Tianshengyuan oral solution was (1400 ± 670) ng. L-1, (n = 10) were also significantly higher than those in the model control group (665 ± 276 ng · L -1, n = 12, P <0.01). In addition, TNF-α decreased to a certain extent. CONCLUSION: Tianshengyuan oral liquid can decrease the production of CD8 + T lymphocytes, increase the ratio of CD4 + / CD8 +, promote the balance of T lymphocyte subsets and promote the production of GM-CSF, Therapeutic effect in model mice.
