目的探讨芥酸酰胺对小鼠的抗焦虑样作用及其机制。方法小鼠每天1次芥酸酰胺灌胃给药,连续7 d。第7天给药后1 h进行高架零迷宫实验或先施行小鼠强迫游泳应激后立即进行水迷宫实验,行为学实验结束后断头迅速取双侧海马备用。分别采用比色法和酶联免疫吸附法(enzyme linked immunosorbent assay,ELISA)测定海马组织单胺氧化酶(monoamine oxidase,MAO)活性、去甲肾上腺素(norepinephrine,NE)和5-羟色胺(5-hydroxytryptamine,5-HT)含量。结果芥酸酰胺显著增加小鼠高架零迷宫和水迷宫实验中的开臂进入次数、开臂滞留时间及缩短寻找目标的潜伏期;降低海马组织中MAO活性,增高NE和5-HT的含量。结论芥酸酰胺对小鼠可能有一定的抗焦虑样作用,其机制可能与调节海马组织单胺类神经递质有关。
Objective To investigate the anti-anxiety-like effect of erucamide on mice and its mechanism. Methods Mice were erucic acid administered orally once a day for 7 days. On the 7th day after the treatment, the elevated zero maze test was performed 1 h after the drug administration or the water maze test was performed immediately after forced swimming swimming in the mouse. After the behavioral test, the bilateral severed hippocampus was rapidly taken. The activities of monoamine oxidase (MAO), norepinephrine (NE) and 5-hydroxytryptamine (5) in hippocampus were determined by colorimetric method and enzyme linked immunosorbent assay (ELISA) -HT) content. Results Erucamide significantly increased the number of open arms in the elevated zero maze test and water maze test, the retention time of open arms and shortened the latency of looking for targets. The activity of MAO in hippocampus and the content of NE and 5-HT were increased. Conclusion Erucamide may have anti-anxiety-like effect on mice, and its mechanism may be related to the modulation of monoamine neurotransmitter in hippocampus.
