球孢白僵菌不同分生孢子特异性表达蛋白分析与筛选

对球孢白僵菌(Beauveria bassiana(Bais.)Vuill)D1-5菌株的3种分生孢子气生孢子、芽生孢子和深层孢子进行电镜观察,并利用双向凝胶电泳技术对3种孢子的特异性表达蛋白进行分析与筛选。结果显示,电镜下气生分生孢子呈球形,外周有棒状结构包裹,具有明显的疏水特性;芽生孢子为圆柱形,大小不均一,表面光滑;深层孢子球形透明,大小均一,表面粗糙,3种分生孢子孢子壁厚度差异不明显。经双向凝胶电泳对3种分生孢子进行特异性表达蛋白分离后,使用PDquest软件对结果进行分析,深层分生孢子与芽生分生孢子蛋白表达图谱相比较,有20个蛋白质表达量发生2倍以上改变,其中8个蛋白质表达量上调,12个蛋白质表达量下调;气生分生孢子与芽生分生孢子蛋白表达图谱相比较,有57个蛋白质表达量发生2倍以上改变,其中43个蛋白质表达量上调,14个蛋白质表达量下调;深层分生孢子与气生分生孢子蛋白表达图谱相比较,有66个蛋白质表达量发生2倍以上改变,其中8个蛋白质表达量上调,58个蛋白质表达量下调。 Three species of conidia, conidia and conidia of conidia of Beauveria bassiana (Bais.) Two kinds of conidial proteins were separated by two-dimensional gel electrophoresis, and the results were analyzed by using PDquest software. Compared with the protein expression profiles of conidia and buds of conidia, 20 proteins were expressed 2 Fold change, of which 8 proteins were up-regulated and 12 proteins were down-regulated. Compared with the protein expression profiles of aerial conidia and buds, 57 proteins changed more than twice, of which 43 Upregulation of protein expression, down-regulation of 14 proteins; compared with the protein expression profiles of the aerial conidia, 66 proteins were more than twice as much, of which 8 were up-regulated, 58 Protein expression decreased.
