目的探讨泡状棘球蚴感染对大鼠牙周炎Th1/Th2平衡的影响。方法 30只Wistar大鼠随机分为泡状棘球蚴感染+牙周炎造模组(棘球蚴+牙周炎组)、牙周炎造模组(牙周炎组)和正常对照组,每组10只。对照组不做任何处理,棘球蚴+牙周炎组和牙周炎组采用结扎线结扎第2磨牙颈部,于丝线上涂抹细菌混悬液,建立牙周炎模型。棘球蚴+牙周炎组建模后经皮肝脏穿刺注射泡状棘球蚴原头节感染棘球蚴。3组分别于造模前、造模后第1、3、6个月采用牙周探针记录实验牙的临床附着丧失和牙周袋探诊深度等变化情况,于造模后第6个月检测大鼠血清和龈沟液白细胞介素-4和干扰素-γ水平,并进行比较。结果棘球蚴感染6个月后,棘球蚴+牙周炎组和牙周炎组临床附着丧失[(3.47±0.48)、(4.38±0.45)mm]、牙周袋探诊深度[(3.53±0.54)、(4.68±0.47)mm]均高于对照组[(0.74±0.29)、(0.74±0.29)mm],牙周炎组高于棘球蚴+牙周炎组(P<0.05);棘球蚴感染6个月后,棘球蚴+牙周炎组和对照组大鼠血清及龈沟液白细胞介素-4水平[(209±71)、(178±56)μg/L,(353±97)、(310±86)μg/L]明显高于牙周炎组[(126±54)、(64±35)μg/L](P<0.05),干扰素-γ水平[(185±53)、(166±45)μg/L,(112±24)、(103±13)μg/L]明显低于牙周炎组[(257±60)、(202±52)μg/L](P<0.05);对照组血清及龈沟液白细胞介素-4水平高于棘球蚴+牙周炎组,干扰素-γ水平低于棘球蚴+牙周炎组(P<0.05)。结论泡状棘球蚴感染使牙周炎大鼠Th1/Th2向Th2偏移,对牙周炎有拮抗作用。
Objective To investigate the effect of alveolar echinococcosis infection on Th1 / Th2 balance in periodontitis in rats. Methods Thirty Wistar rats were randomly divided into two groups: Echinococcus granulosus infection + periodontitis model group (Echinococcus granulosus + periodontitis group), periodontitis model group (periodontitis group) and normal control group . The control group without any treatment, echinococcosis + periodontitis group and periodontitis group ligation of the second molar ligament neck, smear bacterial suspension on the silkworm, the establishment of periodontitis model. Echinococcus granulosus + periodontitis group after injection of percutaneous hepatic puncture of the echinococcosis head infection Echinococcus granulosus. The changes of clinical attachment loss and probing depth of the periodontal pockets were recorded by periodontal probe 1, 3, 6 months after modeling in the 3 groups respectively. At the 6th month after modeling The levels of interleukin-4 and interferon-γ in serum and gingival crevicular fluid of rats were detected and compared. RESULTS: After 6 months of echinococcosis infection, there was a significant loss of clinical attachment between echinococcosis + periodontitis group and periodontitis group (3.47 ± 0.48 vs 4.38 ± 0.45 mm, P < (0.74 ± 0.29) and (0.74 ± 0.29) mm respectively in periodontitis group and periodontitis group (P <0.05) ; The levels of interleukin-4 in serum and gingival crevicular fluid of Echinococcosis + periodontitis group and control group [(209 ± 71), (178 ± 56) μg / L, (35 ± 97) and (310 ± 86) μg / L, respectively, were significantly higher than those in periodontitis group (126 ± 54 and 64 ± 35 μg / L, P < (185 ± 53), (166 ± 45) μg / L, (112 ± 24), (103 ± 13) μg / L] were significantly lower than those in the periodontitis group [(257 ± 60), (202 ± 52) μg / L] (P <0.05). The level of interleukin-4 in serum and gingival crevicular fluid of control group was higher than that of hydatid cyst + periodontitis group, and the level of interferon-γ was lower than that of hydatid cyst + periodontitis group <0.05). Conclusion The infection of Echinococcus granulosus causes the shift of Th1 / Th2 to Th2 in rats with periodontitis, which has an antagonistic effect on periodontitis.
