目的观察银杏叶片对兔动脉粥样硬化过程中血脂代谢及斑块形成的变化,探讨在兔动脉粥样硬化模型中银杏叶片对Connexin43(CX43)表达的影响。方法雄性实验兔40只,随机分为5组,每组8只,对照组普通饮食,其他4组高脂饮食,同时辛伐他丁组给予辛伐他丁灌胃、银杏叶片高低剂量组给予相应剂量的银杏叶片灌胃共12周,检测血脂、主动脉病理观察动脉硬化变化情况,运用免疫组化及Western blot检测主动脉CX43的表达情况。结果银杏叶片组兔的血脂较高脂饮食组下降,动脉粥样硬化的程度较高脂组减轻;CX43的表达较高脂组减少。结论银杏叶片具有轻度的降血脂作用,在一定程度上可以减轻高脂饮食兔动脉粥样硬化程度,这种作用可能是通过下调CX43的表达来实现的。
Objective To observe the changes of blood lipid metabolism and plaque formation during atherosclerosis in rabbits with Ginkgo biloba leaves and to explore the effect of Ginkgo biloba leaves on the expression of connexin43 (CX43) in atherosclerotic rabbits. Methods Forty male rabbits were randomly divided into 5 groups with 8 rats in each group. The rats in the control group were fed with normal diet and the other 4 groups were fed with high fat diet. Simvastatin group was given simvastatin intragastrically and ginkgo leaves were given in high and low dose groups The corresponding dose of Ginkgo biloba for 12 weeks, serum lipids were measured, the changes of arteriosclerosis in the aorta were observed. The expression of CX43 in the aorta was detected by immunohistochemistry and Western blot. Results The blood lipid of Ginkgo biloba group was lower than that of high fat diet group, and the degree of atherosclerosis was lower than that of high fat group. The expression of CX43 was lower than that of high fat group. Conclusion Ginkgo biloba has a mild hypolipidemic effect, which can reduce the degree of atherosclerosis in rabbits to a certain extent. This effect may be achieved through down-regulating the expression of CX43.
