金丝桃素为千层楼(Hypericum perforatum)中的一种活性成分,具有抗抑郁、抗病毒、抗肿瘤的活性,被建议用于AIDS和炎性疾病的治疗。本次研究了金丝桃素对脂多糖(LPS)激活巨噬细胞产生白介素-12(IL-12)的作用及其机制。 结果显示金丝桃素显著抑制LPS诱导IL-12的产生,IC_(50)为1.45μg/mL,而巨噬细胞生存力不变。小鼠RAW264.7细胞瞬时转染试验结果显示金丝桃素强烈抑制IL-12 p40基因启动子的活性,提示金丝桃素对IL-12的产生在转录水平起负调节作
Hypericin, an active ingredient in Hypericum perforatum, has antidepressant, antiviral and antitumor activity and is recommended for the treatment of AIDS and inflammatory diseases. The effect of hypericin on the production of interleukin-12 (IL-12) by macrophages activated by lipopolysaccharide (LPS) was studied in this study. The results showed that hypericin significantly inhibited the production of IL-12 induced by LPS with an IC50 of 1.45 μg/mL, while the macrophage survival was unchanged. Transient transfection experiments in mouse RAW264.7 cells showed that hypericin strongly inhibited the activity of the IL-12 p40 gene promoter, suggesting that hypericin negatively regulates the production of IL-12 at the transcriptional level.
