目的:连续测定犬急性心肌损伤动物模型血清中肌酸激酶MBm ass(CK-MBm ass)、CK-MB活性、CK活性等心肌酶学标志物,观察CK-MBm ass对急性心肌损伤早期诊断的价值。方法:采用球囊堵闭左冠状动脉前降支中远端的方法建立犬急性心肌损伤动物模型。在诸闭前、堵闭后每10m in直至90m in连续采静脉血各2m l,3000 r/m in离心10m in取血清测定或置-40℃保存待测。采用微粒子酶联免疫分析法测定CK-MB-m ass。免疫抑制法测定CK-MB活性,速率法测定CK活性。观察急性心肌损伤后CK-MBm ass、CK-MB活性、CK活性等心肌酶学标志物的动态变化及损伤心肌的组织学改变。结果:冠状动脉堵闭90m in后,心电图可见明显心肌损伤性ST段上抬改变;血清CK-MBm ass、CK-MB活性、CK活性等指标皆超出个体值的2倍以上,但未超出实验参考值的2倍;出现异常值时间CK-MBm ass、CK-MB活性、CK活性分别为(30~45)m in、(30~60)m in、(45~90)m in。结论:CK-MBm ass可早期诊断急性心肌损伤。
OBJECTIVE: To continuously determine the myocardial enzymological markers of creatine kinase MBm ass (CK-MBm ass), CK-MB activity and CK activity in the animal model of acute myocardial injury and to observe the correlation between CK-MBm ass and early diagnosis of acute myocardial injury value. Methods: The animal model of acute myocardial injury in canine was established by occluding the medial and distal ends of left anterior descending coronary artery with balloon. In the closed before the closure of every 10m in until 90m in continuous venous blood collected 2m l, 3000 r / m in centrifuged 10m in serum or set -40 ℃ save the test. CK-MB-m ass was determined by microparticle enzyme immunoassay. CK-MB activity was determined by immunosuppressive assay and CK activity was measured by rate assay. To observe the dynamic changes of myocardial enzymological markers such as CK-MBm ass, CK-MB activity and CK activity after acute myocardial injury and histological changes of injured myocardium. Results: After coronary artery occlusion for 90m in, the obvious myocardial injury ST segment elevation was found on the electrocardiogram. The serum CK-MBm ass, CK-MB activity and CK activity were all more than 2 times higher than the individual value, (30 ~ 45) m in, (30 ~ 60) m in, (45 ~ 90) m in, respectively. The CK-MBm ass, CK- Conclusion: CK-MBm ass early diagnosis of acute myocardial injury.
