目的研究心脑舒通胶囊对蒙古沙鼠脑缺血再灌注(I/R)引发的脑损伤的保护作用。方法按照体重将沙鼠随机分为4组:假手术组、模型组、大小2个剂量(心脑疏通56,14 mg·kg~(-1))实验组,除假手术组外,其余各组采用结扎沙鼠双侧颈总动脉30 min、再灌注6d复制脑缺血模型。每天灌胃给药1次,持续给药7 d。通过HE染色观察海马CA1区神经元细胞的形态变化;免疫组化染色观察海马CA1区胶质纤维酸性蛋白(GFAP)的表达。结果与假手术组存活率(100%)相比,模型组的存活率为66.7%,明显降低,差异有统计学意义(P<0.01);与假手术组神经细胞比例(100.0±16.72)%相比,模型组的神经细胞比例为(45.56±16.72)%,差异有统计学意义(P<0.01);模型组脑组织中海马CA1区神经细胞排列稀疏紊乱且GFAP表达明显增多。与模型组的存活率为66.7%相比,实验组的存活率为86.7%,差异有统计学意义(P<0.01)。与模型组的神经细胞数目为(45.56±16.72)%比较,高剂量实验组细胞的比例为(79.84±15.39)%,差异有统计学意义(P<0.01);同时,心脑疏通组能抑制GFAP表达。结论心脑舒通胶囊能够提高缺血再灌注沙鼠的成活率,改善海马CA1区的神经细胞状态,并能抑制GFAP的表达。
Objective To study the protective effect of Xinnaoshutong Capsules on brain injury induced by cerebral ischemia-reperfusion (I / R) in Mongolian gerbils. Methods According to body weight, gerbils were randomly divided into 4 groups: sham-operation group, model group, 2 doses of size (56,14 mg · kg -1), except the sham-operation group, In the group, bilateral common carotid arteries were ligated in gerbils for 30 minutes, and the cerebral ischemia model was reproduced 6 days after reperfusion. Gavage administered once daily for 7 days. The morphological changes of hippocampal CA1 neurons were observed by HE staining. The expression of glial fibrillary acidic protein (GFAP) in hippocampal CA1 area was observed by immunohistochemistry. Results The survival rate of model group was 66.7%, which was significantly lower than that of sham operation group (100%) (P <0.01). Compared with the sham operation group, the survival rate of the model group was 100.0 ± 16.72% (45.56 ± 16.72)%, the difference was statistically significant (P <0.01). In the model group, the neurons in the hippocampus CA1 area were sparsely arranged and the GFAP expression was significantly increased. Compared with the model group survival rate of 66.7%, the experimental group survival rate was 86.7%, the difference was statistically significant (P <0.01). Compared with the model group (45.56 ± 16.72)%, the proportion of cells in high dose group was (79.84 ± 15.39)%, the difference was statistically significant (P <0.01); at the same time, GFAP expression. Conclusion Xinnaotongtong capsules can increase the survival rate of ischemia-reperfusion gerbils, improve the state of hippocampal CA1 neurons and inhibit the expression of GFAP.
