目的探讨当归补血汤抗动脉粥样硬化(As)损伤作用的机制。方法制备当归补血汤含药血清,台盼蓝染色法检测细胞活性,免疫印迹法检测肿瘤坏死因子-α(TNF-α)、细胞间黏附分子-1(ICAM-1)的蛋白活性变化。结果RAW264.7细胞培养3 d后,台盼蓝染色细胞活力>95%。免疫印迹法检测结果显示,氧化低密度脂蛋白(ox-LDL)刺激RAW264.7细胞后TNF-α蛋白活性明显增强,当归补血汤含药血清组与ox-LDL组比较有差异(P<0.05);ox-LDL刺激RAW264.7细胞后ICAM-1蛋白活性明显增强,当归补血汤含药血清组与ox-LDL组比较有差异(P<0.05)。结论当归补血汤含药血清一定程度上能减少TNF-α、ICAM-1的蛋白表达,减少TNF-α、ICAM-1的表达可能是当归补血汤抗As损伤作用的机制之一。
Objective To investigate the mechanism of Dangguibuxuetang against atherosclerosis (As) injury. Methods The serum of Danggui Buxue decoction was prepared. The cell viability was detected by Trypan blue staining. The protein activities of tumor necrosis factor-α (TNF-α) and intercellular adhesion molecule-1 (ICAM-1) were detected by Western blotting. Results After RAW264.7 cells were cultured for 3 days, trypan blue staining showed> 95% viability. Immunoblotting results showed that the activity of TNF-αprotein in RAW264.7 cells stimulated by ox-LDL was significantly higher than that in ox-LDL group (P <0.05) ). The activity of ICAM-1 protein in RAW264.7 cells stimulated by ox-LDL was significantly increased. There was a significant difference (P <0.05) between Dangguibuxuetang group and ox-LDL group. Conclusion Dangguibuxuetang serum can reduce the expression of TNF-α and ICAM-1 protein and decrease the expression of TNF-α and ICAM-1, which may be one of the mechanisms of Dangguibuxuetang against As injury.
