TRAIL与紫杉醇联合应用对鼻咽癌细胞生长和凋亡的影响

目的:观察肿瘤坏死因子相关凋亡诱导配体(TRAIL)与紫杉醇联合对鼻咽癌CNE-1和CNE-2细胞株的生长抑制作用及凋亡诱导效应。方法:TRAIL和紫杉醇分别单独作用及联合作用于CNE-1和CNE-2细胞后,CCK-8法检测其生长抑制率,流式细胞术检测其凋亡率。结果:TRAIL和紫杉醇对CNE-1和CNE-2细胞均具有抗增殖作用,且在一定范围内呈时间-剂量依赖性;TRAIL与紫杉醇联合应用于CNE-1和CNE-2细胞,其生长抑制率和凋亡率均大于二者单独应用时细胞生长抑制率和凋亡率,差异有统计学意义(P<0.05)。结论:TRAIL和紫杉醇联合作用于鼻咽癌细胞,其生长抑制率和凋亡率显著高于二者单独用药,两药联合应用具有协同杀伤效应,明显优于单独用药。 OBJECTIVE: To observe the growth inhibition and apoptosis-inducing effects of tumor necrosis factor-related apoptosis-inducing ligand (TRAIL) and paclitaxel on CNE-1 and CNE-2 cells. Methods: TRAIL and paclitaxel, alone and in combination, were applied to CNE-1 and CNE-2 cells respectively. The growth inhibition rate was determined by CCK-8 assay and the apoptosis rate was detected by flow cytometry. Results: Both TRAIL and paclitaxel had antiproliferative effects on both CNE-1 and CNE-2 cells in a time-and dose-dependent manner. TRAIL and paclitaxel combined with CNE-1 and CNE-2 cells inhibited the growth of CNE-1 and CNE- The rate of apoptosis and the rate of apoptosis were higher than those of the two alone when the cell growth inhibition rate and apoptosis rate, the difference was statistically significant (P <0.05). CONCLUSION: The combination of TRAIL and paclitaxel inhibits the growth of nasopharyngeal carcinoma cells. The growth inhibition rate and apoptosis rate of TRAIL and paclitaxel are significantly higher than those of the two drugs. The combination of TRAIL and paclitaxel has a synergistic killing effect, which is obviously better than that of single drug.
