目的:观察冰盐水对苯并(a)芘[B(a)P]诱发小鼠前胃癌变过程的影响及其可能的作用机制。方法:昆明种小鼠随机分为4组,分别为正常对照组、B(a)P诱导组、B(a)P加冰盐水诱导组(冰盐水组)和B(a)P加冰水诱导组(冰水组),后3组分别在体内诱导小鼠前胃癌变。比较各组小鼠生存状态以及前胃肿瘤的发生情况;放免法检测各组小鼠第20周正常胃黏膜组织中血栓素B2(TXB2)的表达情况。结果:正常对照组未见肿瘤形成,其余3组均见肿瘤形成,并经病理证实。冰盐水组、冰水组和B(a)P诱导组前胃肿瘤发生率分别为70%、50%和43.33%,冰盐水组成瘤率明显高于冰水组和B(a)P诱导组(P<0.05),而后两者间无显著差异。与正常对照组相比,B(a)P诱导组、冰盐水组和冰水组TXB2表达明显升高(P<0.01或0.05);冰盐水组和冰水组高于B(a)P诱导组(P<0.05),而冰盐水组和冰水组无显著差异。结论:饱和浓度的冰盐水对B(a)P诱导小鼠前胃癌变具有促进作用;其可能通过促进小鼠前胃黏膜TXB2过表达发挥促肿瘤作用。
OBJECTIVE: To observe the effect of ice saline on the process of gastric precancerous lesions induced by benzo (a) pyrene [B (a) P] in mice and its possible mechanism. Methods: Kunming mice were randomly divided into 4 groups: normal control group, B (a) P induction group, B (a) P plus iced saline group (iced saline group) and B Induction group (ice water group), the latter three groups were induced in mice in vivo precancerous lesions. The survival status of the mice in each group and the occurrence of the anterior gastric cancer were compared. The expression of TXB2 in the normal gastric mucosa of the mice in each group was detected by radioimmunoassay. Results: No tumor was found in the normal control group. The tumor formation was observed in the other three groups and confirmed by pathology. The rates of anterior gastric carcinomas in ice-saline group, ice-water group and B (a) P group were 70%, 50% and 43.33%, respectively. (P <0.05), but no significant difference between the latter two. Compared with the normal control group, TXB2 expression was significantly increased in the B (a) P group, the ice saline group and the icy water group (P <0.01 or 0.05), and higher in the ice saline group and the icy water group than in the B (a) P group Group (P <0.05), while there was no significant difference between ice-saline group and ice-water group. CONCLUSION: Ice-salt water with saturated concentration can promote B (a) P-induced precancerous lesions in mice. It may play a role in promoting tumor by promoting TXB2 overexpression in mouse gastric mucosa.
