目的:探讨缺氧诱导因子-2α(HIF-2α)蛋白与妊娠期高血压疾病发生发展的关系。方法:①利用Western blotting和RT-PCR方法分析重度子痫前期患者胎盘组织与正常晚期妊娠胎盘组织中HIF-2α蛋白和mRNA表达的差异。②利用Western blotting和RT-PCR方法分析伴有胎儿生长受限与不伴有胎儿生长受限的重度子痫前期患者胎盘组织中HIF-2α蛋白及mRNA表达的差异。结果:①重度子痫前期组与正常妊娠组比较,胎盘组织中HIF-2αmRNA的表达水平无统计学差异(t=0.86,P>0.05)。②重度子痫前期组与正常妊娠组相比较,胎盘组织中HIF-2α蛋白表达水平明显升高,差异有统计学意义(t=2.45,P<0.05)。③重度子痫前期患者伴有胎儿生长受限组(FGR组)与不伴有胎儿生长受限组(非FGR组)比较,FGR组胎盘组织中HIF-2α蛋白表达水平明显升高(t=3.45,P<0.05),而HIF-2αmRNA表达水平差异无显著性(P>0.05)。结论:HIF-2α蛋白在子痫前期的发生、发展过程中起到一定的作用。重度子痫前期患者胎盘组织中HIF-2α蛋白的高表达可能是导致重度子痫前期患者胎儿生长受限的原因之一。
Objective: To investigate the relationship between the hypoxia inducible factor-2α (HIF-2α) protein and the development of hypertensive disorder complicating pregnancy. Methods: ①The protein and mRNA expression of HIF-2α in placenta of severe preeclampsia and normal placenta of late pregnancy were analyzed by Western blotting and RT-PCR. ② Western blotting and RT-PCR were used to analyze the difference of HIF-2α protein and mRNA expression in placenta between severe pre-eclampsia and fetal growth restriction. Results: ① There was no significant difference in the expression of HIF-2αmRNA in placenta between severe preeclampsia group and normal pregnancy group (t = 0.86, P> 0.05). ② Compared with normal pregnancy group, the expression of HIF-2αprotein in severe preeclampsia group was significantly increased, the difference was statistically significant (t = 2.45, P <0.05). (3) Hypoxia-inducible factor-2alpha protein expression in placental tissue of FGR group was significantly higher than that in FGR group (non-FGR group) (t = 3.45, P <0.05), while there was no significant difference in HIF-2αmRNA expression (P> 0.05). Conclusion: HIF-2α protein play a role in the development of preeclampsia. High expression of HIF-2α protein in placental tissue of patients with severe preeclampsia may be one of the reasons leading to fetal growth restriction in patients with severe preeclampsia.
