目的:观察使用自制小动物定量创伤仪,造成的大鼠全身机械性创伤。方法:首先设计并组装制造实验用小动物定量全身性机械性创伤仪。将40只大鼠分为对照组和创伤组,每组20只(n=20),以40 r/min的转速旋转200圈,分别给予创伤组动物定量创伤,观察心肌组织中心肌细胞的凋亡指数、Caspase-3、iNOS、NO、NADPH氧化酶以及超氧阴离子(.O2-)的表达量,对照组未做任何处理。结果:创伤组心肌细胞的凋亡指数、Caspase-3以及iNOS的表达、NO的产生、NADPH氧化酶的表达及.O2-的生成,均明显高于对照组(P<0.01)。结论:使用自制小动物创伤仪,定量给予大鼠全身性机械性创伤,可造成大鼠心肌细胞发生明显的氧化应激反应和凋亡。
Objective: To observe the use of homemade animal quantitative trauma instrument, resulting in systemic mechanical trauma in rats. Methods: First, the design and assembly of animal experiments used to quantify quantitative mechanical trauma instrument. Forty rats were divided into control group and trauma group, 20 rats in each group (n = 20) were randomly divided into control group and trauma group. The rats were randomly divided into control group and trauma group. The index of death, Caspase-3, iNOS, NO, NADPH oxidase and superoxide anion (.O2-) expression in the control group were not treated. Results: The apoptotic index, the expression of Caspase-3 and iNOS, the production of NO, the expression of NADPH oxidase and the production of O2- in trauma group were significantly higher than those in control group (P <0.01). Conclusion: The use of homemade small animal trauma instrument, quantitatively given systemic mechanical trauma in rats, can cause significant oxidative stress and apoptosis in rat cardiomyocytes.
