目的:分析盐胁迫对菘蓝基因组DNA甲基化模式的影响。方法:采用甲基化敏感扩增多态性(methyla-tion sensitive amplification polymorphism,MSAP)方法。结果:使用15对MSAP选择性引物对盐胁迫前后菘蓝基因组进行甲基化敏感多态性扩增,共得到632条清晰的带纹。共检测到菘蓝基因组44(6.9%)个CCGG位点在盐胁迫后发生了甲基化状态的改变,其中31(4.9%)个位点发生了超甲基化,13(2.0%)个位点发生了去甲基化。结论:盐胁迫能引发菘蓝基因组CCGG位点甲基化模式的改变。
Objective: To analyze the effect of salt stress on methylation patterns of genomic DNA in Taxonomic Tissue. Methods: Methylase sensitive amplification polymorphism (MSAP) method was used. Results: Fifteen pairs of MSAP selective primers were used to amplify methylation-sensitive polymorphisms of the blueberry genomes before and after salt stress. A total of 632 bands were obtained. A total of 44 (6.9%) CCGG loci were found to be methylated after salt stress, of which 31 (4.9%) were hypermethylated and 13 (2.0%) The site demethylation occurred. Conclusion: Salt stress can induce the change of methylation pattern of CCGG locus in Taxus.
