应用溴化乙锭染色的非同位素DNA单链构型多态性分析新技术,对16例卵巢癌及5例正常胎盘组织中P ̄(53)基因的第5、6外显子(Exon5、6)的聚合酶链反应扩增产物进行分析,并结合DNA测序技术进行验证。结果表明正常胎盘组织均未发现异常,3/16例卵巢癌SSCP分析有异常,DNA测序证实这3例基因改变中有2例为核苷酸突变,另1例为核苷酸插入。采用EB染色的非同位素PCR—SSCP,避免了常规SSCP使用放射性同位素的缺点,具有良好的实用性和广泛的应用价值。
A novel technique for non-isotopic DNA single-stranded conformation polymorphism analysis using ethidium bromide staining for exon 5, exon 5 of the P ̄ (53) gene in 16 cases of ovarian cancer and 5 cases of normal placental tissue. 6) Polymerase chain reaction amplification products were analyzed and validated by DNA sequencing techniques. The results showed that no abnormalities were found in normal placenta tissues. 3/16 cases of ovarian cancer had abnormal SSCP analysis. DNA sequencing confirmed that 2 of the 3 gene mutations were nucleotide mutations, and the other one was a nucleotide insertion. The use of non-isotopic PCR-SSCP with EB staining avoids the disadvantages of conventional SSCP using radioisotopes, and has good practicality and wide application value.
