探讨调节性树突状细胞(regulatory dendritic cell,rDC)在移植排斥反应中对T细胞直接识别和间接识别反应的作用。在TGF-β和IL-10的作用下,将小鼠骨髓来源的树突状细胞(DC)培养成rDC,比较rDC和成熟DC在LPS刺激下表型、细胞因子和对同种异体T细胞的刺激活性,检测调节性DC在对受者小鼠的脾脏T细胞直接识别和间接识别的作用。rDC表达共刺激分子明显低于成熟DC,分泌高水平的IL-10,对同种异基因T细胞有明显的抑制作用。在体外中明显抑制了受者小鼠脾脏T细胞的直接和间接识别的反应性增殖。rDC具有明显的异基因抑制作用,对同种异体T细胞的直接识别和间接识别反应性增殖有抑制作用。
To investigate the role of regulatory dendritic cells (rDCs) in the direct and indirect recognition of T cells in transplant rejection. Under the action of TGF-β and IL-10, mouse bone marrow-derived dendritic cells (DCs) were cultured into rDC, and the phenotypes, cytokines and differentiation of rDC and mature DCs stimulated by LPS were compared between allogeneic T cells , The effect of regulatory DCs on direct and indirect recognition of splenic T cells in recipient mice was examined. The expression of costimulatory molecules of rDC was significantly lower than that of mature DCs, secreting high levels of IL-10 and significantly inhibiting allogeneic T cells. Reactive proliferation of direct and indirect recognition of splenic T cells in recipient mice was significantly inhibited in vitro. rDC has significant allogeneic inhibitory effect on allogeneic T cells direct recognition and indirect identification of reactive proliferation inhibited.
