目的 探讨大鼠肝脏Kupffer细胞功能活化与TNT肝毒性的关系及可能机制。方法 运用Kupffer细胞功能活化的大鼠模型 ,观察TNT和VA +TNT联合处理大鼠血清中ALT和AST及肝脏和血清中SOD、CAT和GSH Px活性的变化。结果 TNT组血清中ALT和AST活性明显低于对照组 ,而VA +TNT组则明显高于对照组和相应TNT组。TNT组肝脏和血清中SOD、CAT和GSH Px活性显著和非常显著高于对照组 (P <0 0 5 ,P<0 0 1) ,VA +TNT组某些酶活性显著和非常显著高于对照组和相应的TNT组 (P <0 0 5 ,P <0 0 1)。TNT可加强对Kupffer细胞功能活化大鼠的肝毒性。结论 大鼠肝脏Kupffer细胞经VA活化后 ,可加强TNT肝毒性 ,推测可能与活性氧生成有关。
Objective To investigate the relationship between functional activation of rat liver Kupffer cells and TNT hepatotoxicity and its possible mechanism. Methods The rat model of Kupffer cell activation was used to observe the effects of TNT combined with VA + TNT on the activities of ALT and AST in serum and the activities of SOD, CAT and GSH Px in liver and serum. Results The serum levels of ALT and AST in the TNT group were significantly lower than those in the control group, while those in the VA + TNT group were significantly higher than those in the control group and the corresponding TNT group. The activities of SOD, CAT and GSH Px in liver and serum of TNT group were significantly and very significantly higher than that of the control group (P <0.05, P <0.01), and the activity of some enzymes in VA + TNT group was significantly and very significantly higher than that of the control Group and the corresponding TNT group (P <0.05, P <0.01). TNT potentiates hepatotoxicity in Kupffer cells functionally activated in rats. Conclusion Kupffer cells in rat liver can be activated by VA, which can enhance hepatotoxicity of TNT. It is speculated that this may be related to the generation of reactive oxygen species.
