目的 探讨适用于临床肾移植的组织配型新技术。方法 建立微量序列特异性引物聚含酶链反应(微量SSP法)的方法,对肾移植供、受者SSP-HLA-DRB/DQB 基因分型,并与单克隆抗体一步法分型结果进行对比研究。结果 应用两种方法对142份标本进行HLA-Ⅱ类抗原/基因分型,结果完全相符,微量SSP 法不但快速、需血量少,而且分辨率高、结果准确。结论 微量SSP法具有快速、准确、省血等优点,适合在临床器官移植配型中推广应用。
Objective To explore the new technology of tissue matching suitable for clinical kidney transplantation. Methods SSP-HLA-DRB / DQB genotypes of donor and recipient of kidney transplantation were established by micro-sequence-specific polymerase chain reaction (trace SSP method) and compared with one-step typing of monoclonal antibody the study. Results The HLA-Ⅱ antigen / genotypes of 142 samples were tested by two methods. The results were in good agreement with each other. The trace SSP was not only rapid, but also required less blood, with high resolution and accurate results. Conclusion The micro-SSP method has the advantages of fast, accurate, blood-saving, etc., suitable for clinical application in the matching of organ transplantation.
