目的 :研究肝细胞癌 (HCC)中端粒酶逆转录酶基因hTERT的表达及其与增殖细胞核抗原 (PCNA)和P5 3基因的关系 ,探讨其在HCC发生、发展及预后复发中的意义。方法 :采用免疫组化染色法检测 4 2例HCC组织中hTERT、增殖细胞核抗原(PCNA)和P5 3蛋白的表达 ,并对hTERT与HCC的临床病理特征以及PCNA、P5 3蛋白表达的关系进行分析。结果 :HCC中hTERT、PCNA和P5 3蛋白的阳性率 ,分别为 71.4 % (30 / 4 2 )、76 .2 % (32 / 4 2 )、73.8% (31/ 4 2 ) ;而hTERT在正常肝脏组织中不表达。hTERT在Ⅰ、Ⅱ和Ⅲ级HCC组织中的表达率 ,分别为 4 0 .0 % (4 / 10 )、70 .0 % (14 / 2 0 )及 10 0 % (12 / 12 )。HTERT的表达率与疾病复发有显著相关性 (P <0 .0 5 )。结论 :hTERT表达的异常可能与肝癌的发生、发展有关。hTERT与PCNA和P5 3无明显的相关性。检测hTERT的表达可作为预测肝癌预后复发的潜在指标 ,并提示HCC为由多基因参与的疾病
Objective: To investigate the expression of telomerase reverse transcriptase gene hTERT and its relationship with proliferating cell nuclear antigen (PCNA) and P53 gene in hepatocellular carcinoma (HCC) and its significance in the occurrence, development and prognosis of HCC. Methods: The expressions of hTERT, PCNA and P53 proteins in 42 HCC tissues were detected by immunohistochemical staining. The relationship between hTERT and the clinicopathological features of HCC and the expression of PCNA and P53 protein were analyzed . Results: The positive rates of hTERT, PCNA and P53 protein in HCC were 71.4% (30/42), 76.2% (32/42) and 73.8% (31/42) respectively, while the positive rates of hTERT in normal Not expressed in liver tissue. The expression rates of hTERT in grade I, grade II and grade III HCC tissues were 40.0% (4/10), 70.0% (14/20) and 100% (12/12), respectively. There was a significant correlation between the expression of HTERT and the relapse (P <0.05). Conclusion: The abnormal expression of hTERT may be related to the occurrence and development of liver cancer. hTERT and PCNA and P5 3 no significant correlation. Detection of hTERT expression can be used as a potential indicator of prognosis of HCC recurrence and suggest that HCC is a multi-gene involved disease
