目的:观察扶正化瘀方对实验性肝纤维化的组织形态学影响。方法:用CCl_4诱导Wistar雄性大鼠造成实验性肝纤维化模型,造模同时将其分为模型组、扶正化瘀方组、秋水仙碱组,分别灌胃给药,第6周末处死。肝组织标本作HE染色,用PAP法作Ⅰ、Ⅲ型胶原免疫组化半定量检测,同时测肝组织Hyp含量。结果:与模型组相比,扶正化瘀方组大鼠肝组织结构破坏不明显,纤维结缔组织增生程度轻(P<0.01),Ⅰ、Ⅲ型胶原和肝组织Hyp含量均显著减少(P<0.05,P<0.05,P<0.001),肝功能也有明显的改善。与秋水仙碱组相比,扶正化瘀方组各观察指标的变化似略占优。结论:扶正化瘀方对实验性肝纤维化的形成有显著的抑制作用。
Objective: To observe the effect of Fuzheng Huatan Decoction on the histomorphology of experimental hepatic fibrosis. METHODS: Wistar male rats were induced with CCl_4 to induce experimental hepatic fibrosis model. At the same time, they were divided into model group, Fuzheng Huayu Decoction group and colchicine group, which were administered intragastrically and were sacrificed on the 6th weekend. Hepatic tissue specimens were stained with HE, and PAP method was used for semi-quantitative immunohistochemistry of type I and type III collagen. Hyp content in liver tissue was measured at the same time. RESULTS: Compared with the model group, the structure of the liver tissue of the Fuzheng Huayufang group was not significantly damaged, the degree of fibrous connective tissue proliferation was light (P<0.01), and the contents of type I and III collagen and liver tissue Hyp were significantly reduced (P< 0.05, P <0.05, P <0.001), liver function also improved significantly. Compared with the colchicine group, the Fuzheng Huayufang group’s changes in the observed indicators seemed to dominate. Conclusion: Fuzheng Huayu Decoction has a significant inhibitory effect on the formation of experimental hepatic fibrosis.
