目的观察不同时程吗啡精神依赖大鼠中脑导水管周围灰质(PAG)髓鞘的结构变化。方法将SD大鼠分为吗啡依赖10 d、30 d组、脱毒组及相应的对照组,于大鼠背部皮下注射吗啡,并用条件位置偏爱法观察吗啡精神依赖效应,用电镜观察大鼠的PAG髓鞘结构。结果吗啡依赖10 d组大鼠的PAG髓鞘板层开始松解,扭曲变形,排列较紊乱,部分结构肿胀变性,板层内有空泡;吗啡依赖30 d组和脱毒组大鼠的PAG髓鞘变薄、溶解,呈现典型的脱髓鞘改变;脱毒20 d后未见脱髓鞘改变有修复。结论 PAG髓鞘的损伤呈渐进性,这种损伤可能是阿片类药物成瘾后产生吗啡精神依赖的病理学基础之一。
OBJECTIVE: To observe the structural changes of morphine-dependent long-term myopia of the periaqueductal gray (PAG) in the midbrain of rats. Methods SD rats were divided into morphine dependence 10 d, 30 d group, detoxification group and the corresponding control group. Morphine was injected subcutaneously into the back of rats, and morphine dependence was observed by conditioned place preference. PAG myelin structure. Results In the morphine dependence group, the PAG myelin plate of the rats in the 10-day group began to release, distort, and disordered. Some structures were swollen and degenerated, and there was vacuolization in the lamella. Morphine dependent PAG Myelin thinning, dissolution, showing a typical demyelination change; detoxification after 20 days no demyelination and repair. Conclusions The damage of PAG myelin is gradual, which may be one of the pathological basis of morphine dependence after opioid addiction.
