应用睾丸决定因子(TDF)的基因序列中一对引物F3.2和F4.2,特异地扩增ZFY基因中650bp的DNA片段。其扩增产物在琼脂糖凝胶电泳后,以EB染色直接在紫外透射反射分析仪下观察。本实验能稳定地扩增出500pg男性DNA的特异性电泳带,在混有1:10~2女性DNA的条件下,也能获得预期结果。用此法检测绒毛和羊水各20例,诊断胎儿性别的符合率为100%。
A pair of primers F3.2 and F4.2 in the gene sequence of testis determinant (TDF) was used to specifically amplify a 650 bp DNA fragment in the ZFY gene. The amplified product was subjected to agarose gel electrophoresis and then directly observed under a UV transmission reflectance analyzer by EB staining. This experiment can stably amplify 500pg male DNA specific electrophoresis band, mixed with 1: 10 ~ 2 female DNA conditions, but also to obtain the desired results. Twenty cases of villus and amniotic fluid were detected by this method. The coincidence rate of fetal gender was 100%.
