目的 研究硒对体外神经细胞生长发育的直接影响及可能的作用机理。方法 采用体外新生大鼠皮层神经细胞培养法观察微量元素硒对神经细胞形态学及其表达神经元特异性烯醇化酶 (NSE)和神经生长因子(NGF)蛋白的影响。结果 中低浓度的硒能增加体外培养的神经细胞最长突起长度和细胞平均直径 ;硒能促进体外培养的神经细胞表达 NSE蛋白 ;体外培养的大鼠皮层神经细胞随着培养时间的延长 ,NGF的表达减弱 ,与对照组比较 ,硒能促进神经细胞 NGF的表达。结论 硒可能是通过促进、增强 NSE、NGF蛋白的表达 ,进而直接作用于体外培养的神经细胞 ,促进神经细胞的生长、发育与分化。
Objective To study the direct effect of selenium on the growth and development of neural cells in vitro and its possible mechanism. Methods The effects of trace element selenium on neuronal morphology, expression of neuronal specific enolase (NSE) and nerve growth factor (NGF) in cultured neonatal rat cortical neuronal cultures were observed. Results Selenium at low concentration increased the longest neurite outgrowth length and average cell diameter in vitro. Selenium promoted the expression of NSE protein in cultured neurons in vitro. With the prolongation of culture time, NGF The expression of NGF in nerve cells was promoted by selenium compared with the control group. Conclusions Selenium may promote the expression of NSE and NGF protein and promote the growth, development and differentiation of nerve cells by directly acting on the cultured neurons in vitro.
