目的探讨在中等浓度葡萄糖条件下L-丙氨酸、L-谷氨酰胺、L-亮氨酸、L-精氨酸对INS-1E细胞和小鼠胰岛分泌功能的影响。方法用含有11.1mmol/L葡萄糖和不同氨基酸(AA)的改良Krebs-Ringer缓冲液培养INS-1E细胞和分离的小鼠胰岛,分别测定上清液的胰岛素浓度。结果四种AA均促进葡萄糖诱导的INS-1E细胞和小鼠胰岛的胰岛素分泌,其中同等浓度下L-亮氨酸、L-精氨酸的作用最强。结论四种AA均能增加中等浓度葡萄糖诱导的INS-1E细胞和小鼠胰岛分泌胰岛素。
Objective To investigate the effects of L-alanine, L-glutamine, L-leucine and L-arginine on insulin secretion in INS-1E cells and mice under medium glucose conditions. Methods INS-1E cells and isolated mouse islets were cultured in modified Krebs-Ringer buffer containing 11.1 mmol / L glucose and different amino acids (AA) and the supernatants were each assayed for insulin concentration. Results Four kinds of AA promoted the insulin secretion of insulin-induced INS-1E cells and mouse islets, and the effect of L-leucine and L-arginine was the highest at the same concentration. Conclusion All four AA can increase INS-1E cells induced by medium glucose and insulin secretion from mouse islets.
