目的探讨实验性肝纤维化基质分解素1基因表达的动态变化。方法建立四氯化碳中毒性及人血白蛋白免疫损伤性两种大鼠肝纤维化模型,采用逆转录定量PCR方法,测定肝纤维化不同阶段基质分解素及基因表达水平。结果正常大鼠肝组织有基质分解素1的基因表达。在肝纤维化的发生、发展过程中,肝组织基质分解素1(MMP_3)基因表达逐渐增强;到肝硬化阶段MMP_3基因表达较肝纤维化阶段下降;在肝硬化的自发逆转过程中MMP_3基因表达保持在明显高于正常对照的水平。结论基质分解素1在肝纤维化的发生、发展和逆转过程中可能起重要作用。
Objective To investigate the dynamic changes of gene expression in experimental liver fibrosis. Methods Liver fibrosis induced by carbon tetrachloride (TCC) and immunosuppressive human albumin were established. The levels of stromelysin and gene expression in different stages of hepatic fibrosis were determined by reverse transcription polymerase chain reaction (RT-PCR). Results The normal rat liver tissue had stromelysin 1 gene expression. In the occurrence and development of liver fibrosis, the expression of matrix metalloproteinase-3 (MMP-3) gene in liver tissues gradually increased; the expression of MMP-3 gene in liver cirrhosis stage was lower than that in liver fibrosis stage; in the process of spontaneous reversal of cirrhosis, the expression of MMP- Maintained at a significantly higher level than the normal control. Conclusion Stromaltin 1 may play an important role in the development, progression and reversal of hepatic fibrosis.
