目的:观察加味四物汤降压及对左心室肥厚的作用。方法:将自发性高血压大鼠(SHR)分为模型组、加味四物汤高、中、低剂量组(31.52,15.74,7.81 g.kg-1)及牛黄降压丸(0.17 g.kg-1),卡托普利组(7.88 g.kg-1)。测定用药后各时段的血压变化,并在用药前及8周后分别观察左心室质量指数(LVMI)、心肌胶原含量及Ⅰ,Ⅲ型胶原比值和心肌组织形态学改变。结果:加味四物汤各剂量可使血压不同程度下降,连续给药8周高剂量组血压显示持续降低,与模型组比较有显著性差异(P<0.01)。各剂量组左室质量指数(LVMI)均减小,高剂量组及卡托普利组与模型组比较存在显著差异(P<0.05)。各药物组Ⅰ、Ⅲ胶原比值不同程度的改善。形态学观察显示,与模型组比较,加味四物汤组的心肌纤维结构与分布较好,断裂、坏死和水样变性较轻,胶原沉着较少,毛细血管腔开放,周围渗出较少。结论:加味四物汤具有一定的降压作用,可能对SHR的左心室肥厚有改善作用。
Objective: To observe the antihypertensive effect of Jiawei Siwu Tang and its effect on left ventricular hypertrophy. METHODS: Spontaneously hypertensive rats (SHR) were divided into model group, Jiawei Siwu Decoction high, middle and low dose groups (31.52, 15.74, 7.81 g.kg-1) and Niuhuang Jiangya pill (0.17 g.kg). -1), Captopril group (7.88 g.kg-1). The change of blood pressure at each time after administration was measured. Left ventricular mass index (LVMI), myocardial collagen content, ratio of collagen type I and III, and morphological changes of myocardial tissue were observed before and 8 weeks after treatment. Results: Each dose of Modified Siwu Decoction can reduce the blood pressure to varying degrees. Continuous administration of high-dose 8 weeks high blood pressure group continued to decrease, compared with the model group, there was a significant difference (P <0.01). Left ventricular mass index (LVMI) decreased in each dose group, and there was a significant difference between high dose group and captopril group (P<0.05). The ratio of collagen I and III in each drug group improved to varying degrees. Morphological observations showed that compared with the model group, the Jiawei Siwu Tang group had better myocardial fiber structure and distribution, less fracture, necrosis and watery degeneration, less collagen deposition, capillary lumen opening, and less peripheral leakage. Conclusion: Jiawei Siwu decoction has a certain antihypertensive effect, which may improve the left ventricular hypertrophy of SHR.
