目的:研究椒目油对哮喘大鼠模型Ⅲ型胶原、增殖细胞核抗原(PCNA)的表达水平及气道重建的影响,为椒目的临床应用提供理论依据.方法:将雄性SD大鼠30只随机分为3组:哮喘组、治疗组、对照组,每组动物10只,用卵白蛋白(OVA)腹腔注射致敏和雾化吸入激发建立哮喘大鼠模型,采用计算机图像分析免疫组化气道璧中Ⅲ型胶原的表达,并测量各组支气管壁的厚度.结果:免疫组化结果显示,Ⅲ型胶原阳性表达的平均吸光度:哮喘组大鼠(19.24±0.24)明显高于对照组(2.75±0.22,P<0.01);治疗组(10.16±0.37)明显低于哮喘组(P<0.01).PCNA的表达哮喘组明显高于对照组(P<0.05);治疗组低于哮喘组(P<0.05).哮喘组支气管壁厚度(10.24±2.05)明显高于对照组(5.81±0.73,P<0.01);治疗组(8.45±1.67)明显低于哮喘组(P<0.01).结论:椒目油不但能有效治疗哮喘,而且还能有效防止气道重建,其作用机制可能是抑制了Ⅲ型胶原和PCNA的生成.
OBJECTIVE: To study the effect of peppermint oil on the expression of type III collagen and proliferating cell nuclear antigen (PCNA) and airway remodeling in rat asthma model, and to provide a theoretical basis for the clinical application of pepper. Methods: 30 male SD rats were randomized. They were divided into three groups: asthma group, treatment group, and control group. Ten animals in each group were injected with ovalbumin (OVA) to induce sensitization and nebulization inhalation to establish an asthmatic rat model, and computer image analysis was used to analyze the immunohistochemical airway fistula. The expression of type III collagen was measured and the thickness of the bronchial wall was measured. Results: Immunohistochemistry results showed that the average absorbance of type III collagen positive expression: asthmatic rats (19.24±0.24) was significantly higher than the control group (2.75± 0.22, P<0.01); The treatment group (10.16±0.37) was significantly lower than the asthma group (P<0.01). The expression of PCNA was significantly higher in the asthma group than in the control group (P<0.05); the treatment group was lower than the asthma group (P< 0.05).Bronchial wall thickness (10.24±2.05) in the asthma group was significantly higher than that in the control group (5.81±0.73, P<0.01); the treatment group (8.45±1.67) was significantly lower than that in the asthma group (P<0.01). Oil can not only effectively treat asthma, but also can effectively prevent airway remodeling. Its mechanism of action may be the inhibition of type III collagen. PCNA generation.
