目的观察内源性与外源性不同浓度抵抗素水平对大鼠血清游离脂肪酸(FFA)的影响。方法通过尾静脉注入重组大鼠抵抗素制作外源性高抵抗素大鼠模型,将大鼠分为抵抗素组(RE组,n=8)和生理盐水对照组(NS组,n=8)。饮食诱导并筛选出内源性高抵抗素大鼠模型,将大鼠分为高抵抗素组(HRE组,n=8)、正常抵抗素组(NRE组,n=8)和正常对照组(NC组,n=8),观察不同抵抗素水平下FFA浓度的变化。结果 RE组大鼠血清FFA、胰岛素水平均显著高于NS组(P<0.01)。HRE组大鼠FFA水平较NRE组和NC组均显著升高(P<0.01)。血清抵抗素与FFA、胰岛素、血糖、胰岛素敏感指数(ISI)、体质量、Lee指数均显著相关(P<0.01)。控制胰岛素、血糖、ISI、体质量、Lee指数等混杂因素后,血清抵抗素仍然与FFA显著相关(r=0.615,P<0.01)。结论外源性与内源性抵抗素升高均可引起血清FFA升高;高抵抗素血症在脂代谢紊乱中发挥重要作用。
Objective To observe the effects of different levels of endogenous and exogenous resistin on serum free fatty acid (FFA) in rats. Methods Rat models were made by injecting recombinant rat resistin through caudal vein. The rats were divided into three groups: resistin group (n = 8) and saline control group (n = 8) . The rats were divided into high resistin group (n = 8 in HRE group), normal resistin group (nRE group, n = 8) and normal control group NC group, n = 8). The changes of FFA concentrations at different resistin levels were observed. Results The levels of serum FFA and insulin in RE group were significantly higher than those in NS group (P <0.01). FFA levels in HRE group were significantly higher than those in NRE group and NC group (P <0.01). Serum resistin was significantly correlated with FFA, insulin, blood glucose, insulin sensitivity index (ISI), body weight and Lee index (P <0.01). Serum resistin was still significantly correlated with FFA (r = 0.615, P <0.01) after controlling for confounding factors such as insulin, blood glucose, ISI, body weight and Lee index. Conclusions Both exogenous and endogenous resistin may cause the rise of serum FFA. Hyperinsulinemia may play an important role in the disturbance of lipid metabolism.
