目的:研究脑缺血与B细胞淋巴瘤/白血病-2(B cell lymphoma/leukemia-2,Bcl-2)蛋白的关系及刺五加总黄酮和针刺联合治疗作用。方法:雄性Wistar大鼠线栓大脑中动脉建立脑缺血动物模型,观察大鼠脑缺血后神经功能缺损评分;应用免疫组化法测定模型组(A组)、电针组(B组)、总黄酮组(C组)、针药结合组(D组)、假手术组(E组)、正常对照组(F组)Bcl-2的表达。结果:电针和刺五加总黄酮及其联合治疗组大鼠脑缺血后神经功能缺损评分显著低于单纯缺血组(P<0.01及P<0.05)。与E,F组比较,A,B,C,D组Bcl-2明显增加(P<0.01);与B,C组比较,D组表达明显增多(P<0.01及P<0.05)。结论:刺五加总黄酮和电针能提高Bcl-2蛋白表达,减少脑缺血区细胞凋亡的发生,尤其针药结合治疗有良好的疗效。
Objective: To study the relationship between cerebral ischemia and B cell lymphoma / leukemia-2 (Bcl-2) protein and the combined treatment of total flavonoids and acupuncture of acanthopanax senticosus. Methods: Adult male Wistar rats were established by intracellar middle cerebral artery occlusion (MCAO). Neurological deficit score was observed after cerebral ischemia in rats. Immunohistochemical method was used to detect the changes in model group (group A), EA group (group B) , Total flavonoids (group C), acupuncture combined with drug group (group D), sham operation group (group E) and normal control group (group F). Results: The scores of neurological deficit after electroacupuncture and Acanthopanax flavonoids and their combined treatment group were significantly lower than that of the ischemia group (P <0.01 and P <0.05). Compared with group E and group F, Bcl-2 in group A, B, C and D increased significantly (P <0.01). Compared with group B and C, group B increased significantly (P <0.01 and P <0.05). CONCLUSION: The total flavonoids and electroacupuncture of Acanthopanax senticosus can increase the expression of Bcl-2 protein and reduce the apoptosis of cerebral ischemia. Especially acupuncture combined with acupuncture has a good curative effect.
